Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 1-3x10,000 cells/cm² using 0.05% trypsin/EDTA; 5% CO₂; 37°C. Population doubling is very slow, cells can take up to 5 days to reach 50% confluency; media change 2-3 times a week is recommended with fresh/conditioned medium ratio 1:1 until cell growth is satisfactory.

Culture Medium

Ham's F12 + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 1% Sodium Pyruvate (NaP) + 10% Foetal Bovine Serum (FBS).
