Subculture Routine

Split sub-confluent cultures (70-80%) 1:2 seeding at 2 x 10 4 cells/cm² using 0.05% trypsin/0.025% EDTA in Puck F saline; 2.5% CO₂; 37°C.

Click:RPMI (1:1) + 10µg/ml Insulin + 10mM Hydrocortisone + 5µg/ml Transferrin + 10ng/ml glycyl-L-histydyl-L-lysine acetate + 10ng/ml somatostatin + 10mU/ml Thyroid Stimulating Hormone (TSH) + 2mM Glutamine + 0.1% Foetal Bovine Serum (FBS).
