Cell Line Description

Derived from a 58 year old female Caucasian with Dukes type B adenocarcinoma of the colon. The cells secrete CEA and mucin and are tumorigenic in nude mice. LS174T (ECACC catalogue no. 87060401) was derived from the same patient. The cells have not been exposed to trypsin.

Subculture Routine

On resuscitation, cell viability may be low and cells may take up to 5 days to recover. Split sub-confluent cultures (70-80%) 1:2 to 1:4 i.e., seeding at 2-5x10,000 cells/cm² by scraping. This may also give a reduced viability of the cells before they recover.

Recommended growth conditions: 5% CO2; 37°C.

EMEM (EBSS) + 2mM Glutamine + 1% Non Essential Amino Acids (NEAA) + 10% Foetal Bovine Serum (FBS).
