Cell Line Description

The parental cell line FTC-238 (Cat.No. 94060902) was genetically modified to stably express the human thyroid peroxidase.

Tissue: Thyroid (lung metastasis). Cell line is GMO1 - TPO ORF ligated into pcDNA3 (Invitrogen). Classical transfection/lipofection and selection for G418 resistance and TPO activtiy. Used in a test system for inhibition assays.

Parental line FTC-238 came from a follicular thyroid carcinoma from a 42-year-old male.

The Y chromosome could not be detected in this cell line by short tandem repeat (STR)-PCR analysis when tested at ECACC. It is a known phenomenon that due to the increased genetic instability of cancer cell lines the Y chromosome can be rearranged or lost resulting in lack of detection. The cell line is identical to the source provided by the depositor based on the STR-PCR analysis.

Culture Conditions

Subculture Routine

Split sub-confluent cultures (70-80%), seeding at 0.5-3 x 10,000 cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C.

Culture Medium

IMDM or DMEM:Ham's F12 (1: 1) + 2mM Glutamine + 5% Foetal Bovine Serum (FBS) with the addition of 250µg/ml G418.

At ECACC this was shown to grow in both DMEM and IMDM medium.

Freeze in normal Freeze Mix (10%DMSO+90% FBS).
