Subculture Routine

Resuscitation: Remove cryopreservation medium by dilution in fresh growth medium and pellet cells at 250g and re-suspend cells at 2.5 x10⁵ cells/ml.Subculture the cells between 1-3 x 10⁵ cells/ml. Keep the cell density to less than 1 x 10⁶ cells/ml for optimal growth. Change medium every 2-3 days. Hybridoma growth should double every 24 hours. Loosely attached cells can be pipetted gently from off the plastic.Cryopreserve in either Bambanker (Fisher Scientific, NC9582225), or 90% Foetal Bovine Serum (FBS) + 10% Dimethyl Sulfoxide (DMSO). Growth Conditions: Temperature: 37°C, 5% CO₂: none.

Culture Medium

Excell 610 HSF (Sigma 14610C) + 10% FBS (Gibco 10099141), source-Australia + 1% Glutamax (Gibco 35050).
