Cell Line Description

The continuous cell line My-La CD4+ was established from the independent proliferation of a non-malignant skin homing T cell line from an 82-year old male patient (from a plaque biopsy) with mycosis fungoides stage II by inclusion of IL- 2 and IL-4 in the culture medium.

 

A related cell line was also established from the same patient (My-La Lymphoma, ECACC catalogue No. 95051033).


My-La, CD4+, is also referred to in the literature as My-La, marker chromosome and My-La, Lymphoma is referred to as My-La 46,XY-18+i(1 Bq) or My-La COB+ (ECACC catalogue no. 95051033).


My-La CD4+ is strictly dependent on exogenous IL-2 and IL-4 in the culture medium whereas My-La, Lymphoma on IL-2 only. Both show characteristic chromosome markers and are double homozygous for HLA-A 1.88. My-La, Lymphoma expresses beta-13 of the T cell receptor and phenotypically weak staining was observed for CO2, CD3, COB, CD25, CD71 and TCR-1 alpha/ß.


My-La CD4+ expresses beta-18 of the T cell receptor plus staining for CO2, CD3, CD4,CD25, CD71, HLA-DR and TCR-1 alpha/ B. There has been no evidence of HTLV-1 infection in the cell lines and serum from the patient did not demonstrate antibodies against HTLV-1 and HTLV-11. Both cell lines show a low viability after resuscitation from frozen and a long lag time phase until exponential growth is reached.

Subculture Routine

Maintain cultures between 3-9 x 100,000 cells/ml; 5% CO₂; 37°C. Split only 1:2 as maximum dilution: freeze in 45% growth medium, 45% human AB serum plus 10% DMSO, low viability on resuscitation. Use of human serum produces high levels of debris in cultures. Population doubling time 20-24 hours.

RPMI 1640 + 2mM Glutamine + 10 U/ml IL-2 + 10 U/ml IL-4 + 10% human AB serum
