Cell Line Description

This cisplatin-resistant cell line has been developed by chronic exposure of the parent cisplatin-sensitive A2780 cell line (ECACC catalogue no. 93112519) to increasing concentrations of cisplatin. A2780cis is cross-resistant to melphalan, adriamycin and irradiation. An increased ability to repair DNA damage as well as cytogenetic abnormalities has been observed. In order to retain resistance cisplatinum has to be added to the media for every passage. In addition to this matched pair of drug-sensitive/resistant cell lines an adriamycin-resistant cell line, A2780adr (ECACC catalogue no. 93112520), has been isolated from the same parental line A2780.

Subculture Routine

Split sub-confluent cultures (70-80%) 1:3 to 1:6 i.e. seeding at 2-4 x10,000cells/cm² using 0.05% trypsin or trypsin/EDTA; 5% CO₂; 37°C. Cells will attach slowly after resuscitation and take up to 7 days to reach confluency. Recommendation: resuscitate cells in media without cisplatin. Add after subculture of attached cells.

RPMI 1640 + 2mM Glutamine + 1µM cisplatinum + 10% Foetal Bovine Serum (FBS) .
